CHMOD 444 to mainfile.php fails

I get this error when I click on Admin Index

WARNING: File /../.../../mainfile.php is writeable by the server.
Please change the permission of this file for security reasons.
in Unix (444), in Win32 (read-only)


I chmod it to 444 and it still gives the error and reverts it self back to 644.

I have this problem on my server using FTP to change to that restrictive of a file permission. To get around this I use my server control panel (cPanel) file manager which changes the permissions with no problem.
